Operating Procedures:
Sample Processing:
a) Paraffin Sections:
1. Dewax in xylene for 5–10 minutes.
2. Replace with fresh xylene and dewax again for 5–10 minutes.
3. Immerse in absolute ethanol for 5 minutes.
4. Immerse in 90% ethanol for 2 minutes.
5. Immerse in 70% ethanol for 2 minutes.
6. Rinse with PBS for 2 minutes, repeated 3 times.
7. Stain with Crystal Violet Staining Solution for 10 minutes.
8. Mount the section with Solution C.
Note: If the staining color is too light, extend the staining time. If the staining color is too dark, rinse with Solution B (30 seconds to 1 minute each time), followed by rinsing with water.
b) Frozen Sections:
1. Fix with 4% paraformaldehyde for 30 minutes.
2. Rinse with PBS 3 times, 5 minutes each time.
3. Stain with Crystal Violet Staining Solution for 10 minutes.
4. Mount the section with Solution C.
Note: If the staining color is too light, extend the staining time. If the staining color is too dark, rinse with Solution B (30 seconds to 1 minute each time), followed by rinsing with water.
c) Cultured Cells:
1. Fix with 4% paraformaldehyde for 10 minutes.
2. Rinse with PBS 3 times, 5 minutes each time.
3. Stain with Crystal Violet Staining Solution for 10 minutes.
4. Mount with Solution C.
Note: If the staining color is too light, extend the staining time. If the staining color is too dark, rinse with Solution B (30 seconds to 1 minute each time), followed by rinsing with water.
Precautions:
1. When using this reagent for the first time, it is recommended to take 1-2 samples for a preliminary experiment to determine an optimal staining time.
