Overview

Ethidium Bromide (molecular formula: C₂₁H₂₀BrN₃, molecular weight: 394.31) is a highly sensitive fluorescent dye used for visualizing DNA in agarose and polyacrylamide gels. It emits an orange-red fluorescence signal when excited by a 302 nm UV transilluminator. EB is toxic; all EB-containing solutions must be decontaminated before disposal to prevent environmental pollution.  EB Solution (10 mg/mL) is prepared with sterile double-distilled water and is widely applied in electrophoresis of DNA and RNA samples.

Required Materials:

1. Electrophoresis running buffer (e.g., TAE, TBE)

2. Microwave oven

3. UV analyzer or gel documentation imaging system

Operating Protocol:

1. Melt 50 mL agarose gel solution (commonly 0.8-2% concentration) in a microwave oven.

2. Add 50–100 μL Ethidium Bromide stock solution (10 mg/mL) and mix gently to avoid air bubbles.

3. Pour the gel when cooled to 50–60°C (touchable without scalding). After complete solidification, remove the comb, submerge the gel in electrophoresis buffer, and load samples for electrophoresis.

4. Observe, photograph and document the results under a UV analyzer or gel imaging system.

Precautions:

1. Gel thickness should not exceed 0.5 cm; excessive thickness will reduce detection sensitivity.

2. Avoid prolonged UV exposure when recovering DNA fragments via gel electrophoresis.

3. EB is toxic and irritates the skin and eyes. Handle with extreme caution.

4. For personal safety, wear a lab coat, disposable gloves and a face mask during operation.

5. This product is for scientific research use by qualified professionals only. It must not be used for clinical diagnosis or treatment, food or pharmaceutical production, nor stored in residential premises.
