1. 3×106 Sf9 cells were inoculated into 15 60 mm blood tissue culture dishes containing 5 ml of complete culture medium with 10% fetal bovine serum at 27°C for 2 h to allow the cells to adhere to the wall, and then each time, 7 dishes were infected with wild-type virus and 7 dishes with recombinant virus at an amount of the multiplicity of infections (MOI) of 10, and the remaining one dish was a non-infected control.2. Cells were harvested at different times after infection (from about 15 to 72 h after infection), and non-infected cells were harvested 15 h after infection. Cells and culture supernatants were transferred to centrifuge tubes and centrifuged at 1,000 g at 4°C for 10 min to harvest the cells.3. Process and analyze cells and supernatants (secreted recombinant protein).
