2. Visualization by the naked eye:

The specimen is a transverse section of the esophagus with an irregularly shaped lumen and a complex flat epithelium near the purplish-blue surface of the lumen.

3. low magnification observation:

What is observed in the transverse section of the esophagus is a vertical section of the complex flat epithelium. The complex squamous epithelium and some of the tissue below it are seen to form a protrusion into the lumen of the tube (which is actually a longitudinal fold in a three-dimensional structure). The complex flat epithelium consists of multiple layers of cells, each of which has a different shape. The junction between the epithelium and the deep connective tissue is undulating, separated by a basement membrane. The basement membrane is uniformly pink in color.

4. High magnification observation:

The morphology of the cells in each layer was observed from the basal surface of the epithelium to the luminal surface.

(1) Basal layer: located on the basement membrane, it is a layer of short columnar or cubic cells. The nucleus is deeply stained, ovoid, with little cytoplasm and unclear cell boundaries.

(2) Middle layer: located above the basal layer, composed of several layers of polygonal cells. The nuclei are larger and rounded.

(3) Surface layer: located on the superficial surface of the epithelium, composed of several layers of flat cells. The nuclei are small and pike-shaped.
