Osteoporosis animal model

Summary

Osteoporosis is a common disease of old age, which is mainly characterized by changes in the structure of the bone trabeculae, a decrease in the amount of bone and a weakening of the mechanical strength of the bone. There are many causes of osteoporosis, and the changes in hormone levels in women after menopause are often one of the causes.

Principle

The basic principle of [Osteoporosis Animal Model]: Studies have shown that ovariectomy in female rats can induce a generalized bone loss in rats, and its pathological manifestations are similar to those of postmenopausal bone loss in human females, therefore, it can be used as a very ideal model for the study of postmenopausal osteoporosis.

Materials and Instruments

Material: Female Wistar rats
Reagent: 3% sodium pentobarbital (40 mg/kg)

Move

The modeling method of osteoporosis can be divided into the following steps:
A. Female Wistar rats weighing about 300 g were anesthetized intraperitoneally with 3% sodium pentobarbital (40 mg/kg).
B. Bilateral ovaries were excised by open-abdominal surgery under aseptic conditions.
C. After the operation, the rats were kept in separate cages at room temperature, and were free to ingest water and food.
D. Dual-photon bone mineral density analysis showed a decrease in the amount of bone mineral; histological examination showed a decrease in the number of bone trabeculae and thinning of bone cortex.
