Overview

Combined staining with Hematoxylin and Eosin, abbreviated as HE staining, is the most widely used staining method in pathology and histology. Hematoxylin is a natural basic dye that stains cell nuclei. The main component of chromatin within the nucleus is DNA. In the DNA double helix structure, the phosphate groups on the two nucleotide chains face outward, rendering the outer surface of the DNA double helix negatively charged and acidic. This allows it to readily bind to the positively charged basic dye hematoxylin via ionic or hydrogen bonds, resulting in nuclear staining.

Modified MacConaill Lead Hematoxylin Stain Solution uses lead salts as oxidizing agents and can be used to visualize neuroendocrine cells. This reagent is for research use only and not intended for clinical diagnosis or other applications.

MacConaill Stain Solution: Mix Solution B, Solution C, and Solution D at a ratio of 10:10:1 immediately before use. Prepare and use the solution on-site.
MacConaill Hematoxylin Stain Solution: Mix the prepared MacConaill Stain Solution (BCD mixture) and Reagent E at a ratio of 1:1 thoroughly. Let the mixture stand for 30 minutes and then filter it. Dissolve 3 mL of the filtrate in 15 mL of distilled water. Prepare and use the solution on-site.

Operating Steps (For Reference Only)

1. Deparaffinization of Sections to Water

① Treat sections with xylene or deparaffinization clearing solution twice, 5-10 minutes each time.

② (Optional) Treat with anhydrous ethanol twice, 3-5 minutes each time.

③ 95% ethanol: 3-5 minutes.

④ 90% ethanol: 3-5 minutes.

⑤ 80% ethanol: 3-5 minutes.

⑥ Rinse with tap water or distilled water: 1-3 minutes.

2. Staining

① Differentiate with MacConaill Differentiating Solution for several seconds. For tissues fixed with formalin, paraformaldehyde, or Bouin's solution, differentiate at 60℃ for 3-4 hours; for tissues fixed with glutaraldehyde or Helly's solution, differentiate at 60-65℃ for 12 hours.

② Rinse with distilled water: 5-10 seconds.

③ Pre-warm the prepared MacConaill Hematoxylin Stain Solution to 37-45℃ in advance. Immerse sections in the solution at a constant temperature of 37℃ for 2-3 hours or at 45℃ for 1-2  hours.

④ Rinse with distilled water: 5-10 minutes.

3. Dehydration, Clearing, and Mounting

① 80% ethanol: 10-20 seconds.

② 90% ethanol: 10-20 seconds.

③ Treat with 95% ethanol twice, 1-2 minutes each time.

④ Treat with anhydrous ethanol twice, 2-3 minutes each time.

⑤ Clear with xylene three times, 2-3 minutes each time.

⑥ Mount with neutral resin.

Staining Results

Endocrine cell granules: Dark blue to black

Muscle, nerve, or other tissues: Blue to black

Precautions

1. For tissues fixed with paraformaldehyde or Helly's solution, appropriately extend the staining time.

2. Ensure thorough deparaffinization of sections.

3. Replace the graded ethanol series with fresh solutions regularly.

4. Minimize staining time for frozen sections.

5. For your safety and health, wear a lab coat and disposable gloves during operation.

6. Use the reagent as soon as possible after opening to avoid affecting subsequent experimental results.
