Penicillinase, also known as type I β-lactamase (EC 3.5.2.6), can hydrolyze the β-lactam ring of β-lactam antibiotics, thereby abrogating their antimicrobial activity. This product is a recombinant penicillinase produced by genetic engineering technology. A high-expression engineering strain of penicillinase is constructed, followed by fermentation and purification to obtain a product with high purity and high activity. The product features strong specificity. It can effectively hydrolyze β-lactam antibiotics, including benzylpenicillin (penicillin G), ampicillin, amoxicillin, pivampicillin, carbenicillin, ticarcillin, furbenicillin, mezlocillin, etc.
1. Membrane Filtration Method
① Dilute 1 ml of penicillinase to 1000 ml with sterile water to prepare the washing solution. Sterilize all the equipment to be used by an appropriate method and soak them in the washing solution for 10 minutes.
② Take the prescribed amount of the test sample, filter it according to the specified method, and rinse the filter membrane with an appropriate volume of the washing solution prepared in step ①.
③ Separately dilute 1 ml of penicillinase to 200 ml with sterile water. Add 10 ml of this solution to each culture medium, and store the remaining solution at 2–8 °C for later use.
④ Inoculate the filter membrane into the culture medium according to the specified method.
