Overview
SR 146131 is a potent, orally available, and selective nonpeptide ( cholecystokinin 1 ) receptor agonist.
In Vitro
SR 146131 inhibits in the binding of [ 125 I]-BH-CCK-8S to CCK1sites on 3T3-hCCK1 cell membranes with an IC 50 value of 0.56 ± 0.10 nM. At much higher concentrations, SR 146131 also inhibits the binding of radiolabeled CCK to CCK2sites in CHO-hCCK2 membranes with an IC 50 of 162 ± 27 nM. SR 146131 is a potent CCK1 agonist on several intracellular events linked to CCK1 receptor activation in various cell types: on [Ca 2+ ]i release and IP1 formation, SR 146131 appears as a full CCK1 receptor agonist in the 3T3-hCCK1 cells, but a partial CCK1receptor agonist on MAPK activation and early gene expression in this cell line. SR 146131 also acts as a partial agonist in the two neuroblastoma cell lines. MCE has not independently confirmed the accuracy of these methods. They are for reference only.
In Vivo
SR 146131 completely inhibits gastric and gallbladder emptying in mice (ED 50 of 66 and 2.7 μg/kg p.o., respectively). SR 146131 dose dependently reduces food intake in fasted rats (from 0.1 mg/kg p.o.), in nonfasted rats in which food intake has been highly stimulated by the administration of neuropeptide Y (1–36) (from 0.3 mg/kg p.o.), in fasted gerbils (from 0.1 mg/kg p.o.), and in marmosets maintained on a restricted diet (from 3 mg/kg p.o.). SR 146131 (10 mg/kg p.o.) also increases the number of Fos-positive cells in the hypothalamic paraventricular nucleus of rats. Locomotor activity of mice is reduced by orally administered SR 146131 (from 0.3 mg/kg p.o.) . MCE has not independently confirmed the accuracy of these methods. They are for reference only.

Application Protocols
No tested application protocols are provided for this item.
General practices for small-molecule assay deployment
Stock preparation: Dissolve in anhydrous DMSO to prepare a concentrated stock (e.g., 10–50 mM). Filter if particulate remains (0.22 µm PTFE) and aliquot to minimize freeze–thaw.
Plate-based assays: Normalize final DMSO across wells (typ. 0.1–1.0% v/v). Prepare serial dilutions in DMSO or buffer with co-solvent, mix thoroughly, and pre-wet tips to improve accuracy.
Stability checks: Run time-course UV/LC–MS on working solutions stored at room temperature, 4°C, and assay temperature to rule out degradation.
Data integrity: Include vehicle and background controls; consider counter-screens for fluorescence/absorbance interference.
Note: These are general guidelines, not item-specific instructions. Validate and adapt to your assay and institutional SOPs.
Biological Roles
Item-specific biological target(s), mechanism of action, and pathway roles for SR 146131 are not provided in the Product Data. Do not infer pharmacology without primary references.
General guidance for small-molecule research tools
Define the biological hypothesis a priori and confirm the compound’s identity/purity before drawing mechanistic conclusions.
If literature suggests a receptor/enzyme interaction for SR 146131, validate in your system with concentration–response curves, ligand displacement or activity assays, and appropriate orthogonal methods (e.g., target engagement via CETSA or NanoBRET where applicable).
Assess off-target risks using selectivity panels or predictive in silico profiling (when the structure is available). Counter-evaluate cytotoxicity (e.g., CellTiter-Glo) and nonspecific membrane effects to contextualize activity readouts.
Consider species differences in potency/efficacy if using nonhuman models; lot-to-lot consistency and salt form can shift apparent potency.
Note: This product is supplied strictly for research use only. No medical, diagnostic, or therapeutic applications are intended or implied.
Buffer Applications
This item is a small-molecule research compound, not a buffering agent. It does not define or maintain pH and is not typically used to prepare buffer systems.
Practical notes
When dosing into buffered assays, maintain constant final solvent (e.g., DMSO 0.1–1.0% v/v) across all wells/tubes to avoid pH or activity artifacts.
Verify compatibility of the compound with your buffer salts (e.g., PBS, HEPES, Tris) and additives (detergents, BSA) by observing for precipitation or adsorption losses.
If pH-dependent stability is suspected, perform a small pH-stability scan (e.g., pH 5–9) before large studies.
Green Alternatives
For handling SR 146131 solutions, the primary environmental impacts arise from solvent selection and waste management rather than the compound itself.
Greener solvent considerations (general)
Prefer ethanol or water-based cosolvent systems where compatible with solubility and assay design; they offer lower toxicity and improved biodegradability versus polar aprotic solvents.
If high solvency is required, consider Cyrene or propylene carbonate as greener alternatives to DMF/DMSO in non-biological workflows; confirm compound stability and assay tolerance first.
Comparison (general guidance)
DMSO: Excellent solvency and biocompatibility; moderate environmental profile; ubiquitous in screening.
Ethanol: Renewable routes available; volatile and easy to remove; limited solvency for very lipophilic compounds.
PEG-400 + water: Enhances solubility for hydrophobic molecules; high viscosity can affect liquid handling.
HP-β-cyclodextrin (aq.): Enables aqueous delivery of hydrophobes; biodegradable; may interact with membranes/proteins—assess assay impact.
Operational tips
Minimize total solvent volumes; employ micro-aliquoting and sealed vials to extend stock life.
Consolidate organic waste streams and label by solvent class to facilitate compliant disposal.
Note: Select the greenest option that maintains data quality; verify target readouts are unaffected by the chosen solvent system.
Pharmaceutical Uses
No pharmacopeial status, excipient role, or manufacturing/formulation use is provided for this item.

Practical notes (general guidance)
For small-molecule library members with unknown solubility, begin with DMSO screening (1–30 mg/mL), then evaluate miscibility with water or assay buffer via serial dilution with surfactant (0.01–0.1% Tween-20) when compatible.
If crystalline, gentle warming (25–40°C) and vortex/sonication can aid dissolution; avoid prolonged heating unless thermal stability is documented on the CoA.
Record the exact solvent, concentration, and lot number in assay notebooks to ensure reproducibility across screenings.
Quality and Grades
Grade/Purity: Not specified for this item; refer to CoA/Spec Sheet.
How to interpret grade (general guidance)
Research-grade small molecules typically emphasize identity (1H NMR/LC–MS) and purity by HPLC/UPLC. When UV-Vis chromatograms are provided, verify both area % and the detection wavelength. For bioassays, impurities at even 1–2% can confound SAR—request full analytical package when critical.
If offered as “screening grade,” expect high purity (commonly ≥95% by HPLC, literature) with minimal characterization focused on fit-for-purpose screening; if “analytical” or “reference standard” grade is needed, confirm assay, residual solvents, and water content by Karl Fischer.
Stabilizers and counterions
Stabilizer: Not specified for this item; refer to CoA/Spec Sheet. If present, note its potential interference with bioassays.
Salt form / counterion: Not specified for this item; refer to CoA/Spec Sheet. The salt state strongly affects solubility and pH of stock solutions.
Recommendations
Request: detailed CoA (HPLC trace, MS, NMR), salt form, residual solvent profile, water content, and any chirality data. Align analytical controls with your intended application (e.g., GLP screening vs. discovery assays).
Reaction and Applications
This product is positioned as a small-molecule library member rather than a synthetic reagent. No manufacturer application notes are provided for chemical reactions.
Research applications (general)
Reference compound in high-throughput screening (HTS), hit validation, and mechanistic assays where SR 146131 is known/assumed to interact with a biological target in your assay system. Confirm exact target and potency from primary literature before use.
Tool compound usage in cellular or biochemical assays to probe pathway dependence, receptor occupancy, or as a positive/negative control once target identity is confirmed.
Practical guidance
Prepare concentrated DMSO stocks (see Solvent Selection) and dispense using low-retention tips. For plate-based assays, equalize final DMSO across wells to avoid solvent bias.
If used for SAR benchmarking, run orthogonal readouts (e.g., biochemical vs. cell-based) to control for assay interference (fluorescence quenching, redox activity, aggregation).
Implement counter-screens (e.g., detergent-sensitive aggregation tests, redox-cycling detection with HRP/resazurin) to rule out pan-assay interference compounds (PAINS) behavior.
Note: No specific reaction chemistry (e.g., cross-coupling, condensation) is applicable here given the lack of structural and functional group disclosure.
Reaction Conditions
Not applicable. This product is not positioned as a chemical reagent for synthesis, and no item-specific reaction conditions are provided.
If employing derivatization for analytical purposes (general)
Conjugation or tagging steps (e.g., forming reporter-labeled analogs) should be designed only after the exact structure is confirmed. Optimize solvent, temperature, and base/acid conditions around the known functional groups.
Maintain an analytical baseline (LC–MS, HPLC purity) before and after any modification to confirm integrity and absence of degradation byproducts.

General laboratory precautions (small-molecule screening compounds)
PPE: Lab coat, safety glasses, and appropriate chemical-resistant gloves (e.g., nitrile). Avoid inhalation of dusts/aerosols. Handle powders in a fume hood or ventilated enclosure.
Storage incompatibilities: Until specific data are known, segregate from strong oxidizers, strong acids/bases, and reducing agents. Protect from light and moisture unless stability data indicate otherwise.
Hygiene: Wash hands after handling; do not eat, drink, or smoke in work areas. Prevent environmental release; dispose of waste per institutional and local regulations.
First aid (overview; see SDS): Eye/skin contact—rinse with water for ≥15 minutes; remove contaminated clothing. Inhalation—move to fresh air. Ingestion—rinse mouth; seek medical attention. Provide SDS to responders.
Weighing and solution prep: For microgram–milligram quantities, use anti-static precautions and weigh boats/lined spatulas to minimize loss and exposure.
Note: Many heteroaromatic or halogenated research compounds can be harmful if swallowed or inhaled; adopt a conservative risk posture in the absence of detailed hazard data.
Solvent Selection
Without a disclosed structure for SR 146131, choose solvents using a tiered, assay-compatible approach.
Primary stock solvent
DMSO: Default for discovery libraries due to broad solvency and assay compatibility. Start at 10–50 mM stocks. Filter-sterilize when required (0.22 µm PTFE). Verify assay tolerance (typically ≤0.5–1.0% v/v).
Secondary organic cosolvents (if DMSO is insufficient)
N,N-Dimethylformamide (DMF) or N-methyl-2-pyrrolidone (NMP): High solvency; confirm assay tolerance and mitigate leachables from plastics (prefer glass vials).
Ethanol or methanol: Useful for less polar compounds with better aqueous dilution behavior than pure DMSO in some systems.
Aqueous handling
Prepare aqueous working solutions by stepwise dilution of DMSO stocks into buffer under vortexing. Include surfactant (e.g., 0.01–0.05% Tween-20) if compatible to reduce precipitation.
For poorly soluble compounds, consider co-solvent systems (e.g., 2–10% DMSO + 10–20% PEG-400 in buffer) or cyclodextrin inclusion (e.g., 5–20% w/v HP-β-CD), pending assay constraints.
Material compatibility and stability
Prefer borosilicate glass for concentrated stocks; minimize freeze–thaw.
Evaluate photostability (wrap in foil) and hydrolytic stability (pH 2–9 screening) empirically if no data are provided.
Documentation
Record solvent, concentration, pH, and any visible precipitation after dilution. Retain aliquots for stability tracking.
