Overview

This product is used to terminate enzymatic digestion reactions of tissues or cells, rapidly neutralize enzyme activity, and protect cell integrity. It is widely used in the preparation of single‑cell suspensions.It can stop enzymatic digestion of various animal organs (including brain, liver, spleen, kidney, lung, tumor tissue, etc.) mediated by tissue single‑cell dissociation enzymes, thereby protecting tissue cells from damage caused by excessive enzymatic digestion.This greatly improves the success rate and data quality of downstream experiments such as flow cytometry, single‑cell sequencing, and cell culture.


Protocol

  1. After completion of enzymatic digestion, add an equal volume or 2× volume of Stop Solution to the digestion system immediately (adjust the volume according to enzyme concentration and reaction conditions).
  2. Mix gently and incubate at room temperature for 1–2 minutes to fully terminate enzyme activity.
  3. Centrifuge at 300–400 ×g for 5 minutes to collect cells; discard the supernatant.
  4. Resuspend the cell pellet in pre‑chilled PBS or medium for downstream experiments including flow cytometry, single‑cell sequencing, and cell culture.

Notes

  1. Mix the Stop Solution thoroughly before use. If precipitation occurs, centrifuge and use the supernatant.
  2. Enzymatic digestion conditions vary among tissue types. Pre‑experiments are recommended to optimize the volume of Stop Solution and reaction time.
  3. This product is sterile. Use aseptic technique to avoid contamination.
  4. For research use only.
