Carnoy's Fluid is widely used for the visualization of DNA and RNA, as well as the fixation of glycogen and Nissl bodies. It features extremely rapid penetration, and the fixation duration shall not exceed 24 hours. This fixative can quickly infiltrate cells to preserve the integrity of chromosome structure, and enhance the basophilia of chromosomes for optimal staining results. It is recommended for tissue fixation prior to RNA, DNA and glycogen staining, and can render glycogen into fine granular forms. Thanks to its strong penetrability, it is also suitable for tissues with dense outer membranes that are difficult for common fixatives to infiltrate. Fixed tissues can be temporarily stored in 70% ethanol if not used immediately.
Instructions for Use:
1. Fix small tissue pieces for 30 to 40 minutes.
2. Fix medium-sized tissue pieces for 2 to 4 hours.
3. Prolong the fixation time appropriately for large tissue blocks, but do not exceed 24 hours.
Precautions:
1. Carnoy's Fluid is harmful to the human body. Handle with caution. It is not suitable for tissues intended for lipid staining.
2. Fixation time varies depending on the thickness of tissue specimens.
3. Use a sufficient volume of fixative. The volume ratio of fixative to tissue block should be greater than 10:1.
4. Temperature exerts a notable effect on fixation. Elevating temperature accelerates the fixation process, yet excessive temperature should be avoided.
5. Fresh tissues must be fixed promptly. If immediate fixation is not available, preserve the tissues in normal saline and deliver for processing in a timely manner.
6. Wear a lab coat and disposable gloves during operation for personal safety and health protection.
