L'azodicarboxylate de diéthyle agit comme un diénophile utilisé dans les réactions de Diels-Alder. Il est utilisé comme réactif dans l'alfa-thiocyanation des cétones énolisables avec le thiocyanate d'ammonium et dans l'annulation des imines protégées par un N. Il sert de réactif pour la préparation d'immunostimulants alfa-Galactosylcéramides, d'inhibiteurs de bisubstrats avec reconnaissance moléculaire sur le site actif de la catéchol-O-méthyltransférase et d'aza-bêta-lactames via la cycloaddition [2 + 2] catalysée par le NHC avec des cétènes. Il trouve une application dans la réaction de Mitsunobu et est utilisé dans la synthèse de produits pharmaceutiques tels que la zidovudine et la procarbazine.
