Experiments on the oxygen requirement for rice seed germination

Summary

Through the experiment, it is proved that the normal germination and growth of seeds of higher plants must have sufficient oxygen, even for flooded plants such as rice.

Operation method

Experiments on the oxygen requirement for rice seed germination

Principle

Rice is a submerged plant, and has a certain ability to adapt to the oxygen-deficient environment, but the way of adaptation is mainly with the help of well-developed aeration organization so that the root system gets the oxygen transported from the ground. Rice seeds can germinate in water with insufficient oxygen, which is not possible for general crops such as wheat and corn. However, under severe oxygen deprivation, its embryonic roots and true leaves cannot grow normally either, and only slender shoot sheaths can grow. As the shoot sheath quickly extends out of the water, it can transport the oxygen in the air to the base, which is conducive to the development of roots and leaves.

Materials and Instruments

Rice seeds
Mercury solution
Light incubator 500 ml beaker Pointed tweezers Plastic ruler

Move

1. Seed treatment

Full rice seeds were selected and surface sterilized with 0.1 % mercuric chloride solution for 10 min, then the seeds were washed well with water and soaked for a few hours.

2. Seed culture

Take five 500 ml beakers, numbered and labeled for five treatments, put 30-40 seeds in each treatment (beaker), then add different depths of distilled water to each treatment as follows.

1. Just add water to half the thickness of the seeds.

2. Add water to submerge the seeds for 1cm.

3. Add water to submerge seeds for 3cm.

4. Add water to submerge seeds for 5cm.

5. Add water to submerge seeds for 7cm.

The treatments were incubated in a bright place at 25-30°C for 5-7d (during which time the water was changed once in 2 days).
