1. Staining: HE
2. Visualization by the naked eye:
The specimen is a contracted bladder, and the purplish-blue side is the metaplastic epithelium on the luminal surface of the bladder.
3. low magnification:
The metaplastic epithelium consists of multiple layers of cells, each with different morphology. The free surface of the epithelium is basically parallel to the basal surface, and the basement membrane is not obvious.
4. High magnification observation:
Observe the morphology of the cells in each layer from the deep side to the shallow side.
(1) Basal layer: a layer of dwarf columnar cells.
(2) Middle cell layer: located above the basal layer, with several layers of irregular polygonal cells.
(3) Surface cells: located on the surface of the epithelium, a layer of rectangular or cuboidal cells, with large cells, sometimes with two nuclei inside the cell. The cytoplasm near the surface stains dark red.
