This Lentiviral Solution Buffer adopts an optimized formulation based on the HEPES‑sodium bicarbonate buffering system, supplemented with L‑glutamine, trace elements and cryoprotectants. It is used for resuspending lentiviral pellets obtained from ultracentrifugation, PEG precipitation and ultrafiltration concentration. Lentiviral particles resuspended in this buffer show low cytotoxicity, good storage stability and tolerance to multiple freeze‑thaw cycles, and are suitable for various lentivirus‑related experiments.
Instructions for Use
Resuspension of virus pellets after ultracentrifugation or PEG precipitation
1. Add an appropriate volume of Lentiviral Solution Buffer according to the expected concentration factor (e.g., for 100 mL original lentivirus stock expected to be concentrated 100‑fold, add 1 mL of Lentiviral Solution Buffer), then gently pipette up and down to resuspend the pellet.
2. Allow the mixture to stand for 4‑6 h to enable lentiviral particles in the pellet to be released into the Lentiviral Solution Buffer.
3. Centrifuge at 11,200 × g (~12,000 rpm*) for 3 min at 4 °C. Collect the supernatant, aliquot it into suitable volumes and store at −80 °C.
*Conversion for small‑volume centrifuge with approximately 7 cm effective rotor radius (for ≤2 mL centrifuge tubes).
Buffer exchange after ultrafiltration concentration
After ultrafiltration of the original lentivirus stock, add an appropriate amount of Lentiviral Solution Buffer, perform centrifugation until the target volume is reached, aliquot and store at −80 °C.
