This method is based on the principle of PAS‑NYS dual staining. Periodic acid selectively oxidizes vicinal diol groups in polysaccharide structures to aldehyde groups, which subsequently react with colorless fuchsin (Schiff's reagent) to form a stable magenta product, thereby localizing polysaccharides. Meanwhile, Naphthol Yellow S, an acidic dye, specifically binds to proteins rich in basic amino acids under acidic conditions, imparting a yellow coloration. The combination of these two staining reactions enables the simultaneous comparative visualization of polysaccharides and basic proteins on the same section.
