Vue d’ensemble

Product Characteristics

UNI-StabilPLUS is a universal stabilizer for the dilution and stabilization of both Horseradish Peroxidase (HRP) and Alkaline Phosphatase (AP) labeled proteins and antibodies, in order to maintain the molecular conformation and prevent loss of activity over time. This enables the making of pre-diluted, ready-to-use conjugates, minimizing assay errors in dilution.

Superior stabilization of HRP and AP conjugated antibodies in low as well as high protein dilutions is seen, when using UNI-StabilPLUS.

When tested with AP conjugated antibody stability is seen as follows:

• at least 3 years at 2-8 °C

• at least 2 years at room temperature

• at least 4 weeks at 37 °C

When tested with HRP conjugated antibody stability is seen as follows:

• at least 2 years at 2-8 °C

• at least 1 years at room temperature

• at least 2 weeks at 37 °C

UNI-StabilPLUS is recommended for the dilution of antibodies directed against rabbit immunoglobulins unlike HRP-StabilPLUS (cat. no. H494387) and Antibody Enhancer (cat. no. A494276).

Composition & Properties

UNI-StabilPLUS is a ready-to use buffer that appears as an opaque solution. The product is based on a mild acid Tris buffer containing proprietary stabilizing components. UNI-StabilPLUS contains neither BSA, nor other material from bovine serum, no azide, mercury or other toxic components.

Working Procedure

1.Make a series of dilutions of the HRP- or AP conjugated protein in UNI-StabilPLUS in order to determine the optimal dilution.

2.Run the assay as usual or store the diluted conjugated protein preferably at 2-8 °C.

Tips & Tricks

• Avoid using phosphate buffers for AP-conjugated antibody assays. We recommend the use of Tris/HCl, Tween as the washing buffer, instead of a PBS buffer which will reduce signal significantly.

• For extended stability of HRP conjugated antibodies, HRP-StabilPLUS (cat. no. H494387) is recommended.

Handling & Storage

• Store solution at 2-8 °C.

Application Protocols

Item-specific, validated protocols are not provided in the Product Data. The following are general, non-product-specific workflows for using antibody stabilizers.

  • ELISA (general)

    • Prepare coating, blocking, and wash buffers as usual. Dilute detection antibodies in buffer containing a trial level of stabilizer (e.g., 2–5% v/v; not product-specific). Compare signal/background versus control without stabilizer. Verify compatibility with HRP/AP.
  • Western blot (general)

    • Prepare primary/secondary antibodies in TBST or PBST supplemented with stabilizer at low percentage to reduce nonspecific adsorption to plastics. Avoid high excipient loads that may affect membrane interactions.
  • IHC/IF (general)

    • For working dilutions, include minimal stabilizer necessary to maintain signal over the incubation period. Protect from light when using fluorophore-conjugates. Validate that excipients do not interfere with antigen retrieval or mounting media.
  • Flow cytometry (general)

    • Add stabilizer only to antibody master mix, not to cell suspensions, unless validated to avoid effects on cell membranes. Keep samples cold and protected from light.
  • Storage of diluted antibodies (general)

    • Short-term (hours–days) at 2–8°C with stabilizer. For longer storage, aliquot and freeze only if SDS/CoA indicates compatibility. Avoid repeated freeze–thaw.

For UNI-StabilPLUS-specific instructions (concentrate/ready-to-use status, recommended ratios), consult the CoA/Spec Sheet.

Biological Roles

Not typically applicable. UNI-StabilPLUS is a laboratory stabilizer mixture rather than a defined biomolecule with a physiological role. If any biological additives are present (e.g., proteins or amino acids), those roles pertain to formulation performance, not endogenous biology. For composition and any biological-source components, consult the CoA/SDS.

Buffer Applications

Relevance

  • High relevance. UNI-StabilPLUS is intended for antibody/protein stabilization within buffer systems. Exact composition, pH, and recommended dilution are not specified here; consult CoA/Spec Sheet.

General best practices (literature/general)

  • Working dilutions: Prepare antibody dilutions in PBS or TBS containing the stabilizer at the vendor-recommended ratio. If unavailable, pilot a small range (e.g., 1–10% v/v stabilizer) and select the lowest concentration that maintains performance.
  • Blocking and background: Stabilizers sometimes reduce nonspecific adsorption. If blocking is still required, use BSA or casein in a separate step; verify compatibility.
  • Adsorption control: Use low-bind plastics and include stabilizer to minimize loss of low-abundance antibodies.
  • Freeze–thaw mitigation: Combine stabilizer with aliquoting; avoid repeated freeze–thaw where possible. Confirm whether the stabilizer is intended for cold storage or only for short-term, 2–8°C storage.
  • Enzyme/fluorophore compatibility: Ensure that excipients do not inhibit HRP/AP or quench fluorophores. Run small controls.

Example workflow (generic)

  1. Equilibrate antibody to 2–8°C.
  2. Prepare buffer (PBS/TBS) at working ionic strength.
  3. Add stabilizer to buffer at trial ratios (e.g., 2%, 5%, 10% v/v; not product-specific), mix gently.
  4. Dilute antibody to working concentration; incubate briefly and proceed with assay or store short-term at 2–8°C protected from light.
  5. Record performance metrics (signal, background) to optimize the ratio.

Note: For exact usage instructions unique to UNI-StabilPLUS, refer to CoA/SDS.

Green Alternatives

Context

  • As an aqueous laboratory stabilizer, UNI-StabilPLUS is generally aligned with low-volatility, nonflammable reagents. However, environmental profile depends on undisclosed formulation specifics.

General greener-practice guidance (literature/general)

  • Prefer aqueous, preservative-optimized solutions that minimize hazardous biocides where possible.
  • Use smallest effective volumes to reduce waste; adopt aliquoting strategies to extend antibody life without repeated freeze–thaw.
  • Choose low-toxicity surfactants and readily biodegradable excipients when alternatives are available (verify composition on CoA/SDS).
  • Implement closed, reusable cold-chain packs for shipping/storage to reduce single-use materials.

Comparison table (general, not product-specific)

  • Aqueous stabilizer vs. glycerol-heavy mixes
    • Benefits of aqueous: lower viscosity, better pipetting, often lower environmental impact.
    • Tradeoffs: may provide less freeze protection than high-glycerol mixes; composition for UNI-StabilPLUS is not specified.

Disposal

  • Treat spent solutions as aqueous laboratory chemical waste unless your SDS indicates otherwise. Avoid drain disposal if preservatives (e.g., azide, isothiazolinones) are present; confirm on SDS.
Pharmaceutical Uses

Not applicable for therapeutic or clinical use. UNI-StabilPLUS is designated For research use only and is intended for laboratory stabilization of antibody/protein reagents. No pharmacopeial status, excipient listing, or GMP manufacturing details are provided. Do not use in human or veterinary applications.
