This product is an exosome qualitative and quantitative detection kit based on the sandwich ELISA method. It adopts high-performance CD9 capture antibody and CD81 detection antibody to sensitively detect CD9 and CD81 targets co-expressed on the surface of exosomes. Compatible with various samples such as human body fluids and cell culture media, this kit enables rapid and accurate quantification of exosomes in raw samples.

Product Principle:

Test Procedure:
1. Pre-test Preparation
① All reagents shall be equilibrated to room temperature for at least 15 minutes before use.
② Dilute the 20× Concentrated Wash Buffer with distilled or deionized water to prepare 1× working solution, mix thoroughly for later use. (The kit is supplied with assay diluent, which can be used to dilute high-concentration standards as required.)
2. Operating Steps
① Take the equilibrated microplate strips and reagents, and fully shake all reagents evenly.
② Add serially diluted Exosome Standards and test samples into respective wells of the microplate (100 μL per well).
③ Seal the reaction wells with plate sealer, and incubate at a constant temperature of 37 °C away from light for 60 min.
④ After incubation, wash wells with wash working solution 3 times, 200–250 μL per well each time. Blot dry on absorbent filter paper after washing.
⑤ Add 100 μL Enzyme Conjugate to each well, seal the plate, and incubate at 37 °C in the dark for 60 min.
⑥ Repeat the washing procedure 3 times as described above, then blot dry completely.
⑦ Prepare chromogenic substrate working solution: mix Chromogenic Solution A and Solution B at an equal volume ratio. Use the mixed substrate within 2 minutes after preparation.
⑧ Pipette 100 μL freshly prepared chromogenic substrate working solution into each well, and incubate at 37 °C protected from light for 30 min.
⑨ Add 50 μL Stop Solution to each well, mix gently, and measure OD₄₅₀ values immediately (within 3 min). Plot an XY linear calibration curve.
3. Calibration Protocol
XY linear fitting is adopted, with a mandatory requirement that the coefficient of determination R² ≥ 0.99. A full set of calibrants must be tested in each independent experiment to generate the calibration curve.
