Experimental morphological observations on vesicles in young wheat roots

Summary

The tip of the germinating wheat embryonic root was cut (about 1 cm from the tip) and immersed in neutral red solution for live staining for 10-15 min, then removed and rinsed with tap water for 5-10 min, and the epidermis was torn (or made a longitudinal section of the severed root) and examined under a microscope.

I. Materials and equipment

Sprouted wheat radicle, microscope, carrier sheet, cover sheet, forceps, dropper, razor blade, 0.03% neutral red solution, small petri dish.

II. Experimental steps

Cut the tip of sprouted wheat radicle (about 1cm from the tip), immerse it in neutral red solution for 10-15 minutes, then remove it and rinse it with tap water for 5-10 minutes, tear the epidermis (or make a longitudinal section of the root) and examine it under the microscope. It can be seen: in the meristematic zone embryonic cells there are a small number of sporadic small red dots, indicating that the vesicle has begun to form; into the elongation of the cell there is a red reticulated interlacing vesicle structure; in the mature section of the cell, there is a very complete large vesicle, generally located in the center of the cell.
