Principle

The amount of wounding is an important indicator of plant root vigor. When the amount of wounding is high, the volume can usually be measured directly, while for some plants with less wounding, the amount can be determined by weighing the plant after absorption with skimmed cotton.

Materials and Instruments

Finger tubes Soldering iron Cellophane Degreased cotton Cotton wool Balance

Move

1. Fill the finger tube with skimmed cotton, loosened and tightened appropriately, and weigh it on a balance together with a piece of tied thread and record the weight.

2. Cut the above-ground part of the plant to be tested at 2-3cm from the ground with a sharp knife, and then immediately put the finger-shaped tube prepared above on the sleeve, and tie it tightly with a piece of thread so as not to evaporate the wound fluid (note that it is important to make sure that the skimmed cotton in the sleeve is in close contact with the incision of the plant).

3. After a period of time, remove the casing, tie the open end with cotton thread and weigh it on the balance, determine the difference between before and after weighing as the wound flow rate, and find out the wound flow rate per unit of time.
