Technical Principle

Protocol
- Sample Preparation: Resuspend the dissociated single-cell suspension in PBS to a density of 1–5×10⁶ cells/mL, and mix with the debris removal reagent at a 1:1 volume ratio. If debris is abundant, increase the proportion of the debris removal reagent.
- Centrifugation: Centrifuge at 400–800×g for 10 minutes (enable full acceleration and deceleration of the centrifuge). After centrifugation, debris is enriched in the upper layer, while intact cells are pelleted at the bottom.
- Cell Collection: Carefully aspirate and discard the supernatant, then resuspend the cell pellet in PBS.
- Washing and Resuspension: Wash 1–2 times with PBS or culture medium to obtain high-purity, high-viability target cells for downstream experiments.
Notes
Debris levels may vary depending on the tissue and cell type. For optimal debris removal efficiency, pre-experiments can be performed to adjust the volume mixing ratio of the debris removal reagent to the cell suspension for better results.
