Overview

Hydroxyproline (4‑hydroxyproline, Hyp) is a non‑essential amino acid and one of the major components of collagen. In mammals, Hyp is found almost exclusively in collagen and elastin, whereas in plants, it is widely distributed in various proteins. Numerous diseases associated with abnormal collagen metabolism can lead to alterations in Hyp levels in blood, urine, and tissues. Therefore, Hyp level is an important reference indicator for the clinical evaluation of collagen metabolism‑related disorders. This kit employs a colorimetric method for the determination of Hyp content. After acid hydrolysis, free Hyp is released from the sample. The free Hyp is oxidized by Chloramine T, and the oxidation product reacts in the presence of p‑dimethylaminobenzaldehyde (DMAB) to form a purple‑red compound, which exhibits a characteristic absorption peak at 560 nm. By measuring the absorbance of the reaction solution at 560 nm (which is directly proportional to the Hyp content), the hydroxyproline concentration in the sample can be quantitatively calculated.
