This product is a ready to use, latest generation efficient and rapid blocking solution for Western Blot or ELISA experiments. It uses a protein free formula and has a significantly better overall effect than traditional blocking solutions based on BSA (bovine serum albumin), skim milk powder, casein (Casein), and similar foreign products. This reagent does not contain protein components of mammalian origin and has no endogenous phosphorylated proteins or biotin. It also does not interfere with phosphorylated specific antibodies and biotin.
Precautions:
1. The blocking time of different antibodies varies, and can be operated according to specific experiments. For antibodies with relatively high background values, the blocking time can be appropriately extended to 30-60 minutes. If there is a special need, it can also be left overnight at 4 ° C.
2. There is no single sealing solution suitable for all experimental systems, so for some special experiments and antibodies, it may be necessary to consider whether to replace them with other sealing agents, such as BSA or skim milk powder, depending on the specific situation.
3. This product is for scientific research purposes only and should not be used for any other purposes.
Instructions for Use:
1. After the membrane transfer is completed, wash the protein membrane with Western wash solution for about 1-5 minutes.
2. According to the size of the membrane, add the sealing solution to a petri dish or other suitable container to ensure that the sealing solution can completely cover the membrane. For conventional Western blotting, it is recommended to use about 10ml of sealing solution for a 7.5 × 8cm membrane.
3. Gently clamp the membrane into the sealing solution using flat tweezers, ensuring that the sealing solution completely covers the membrane. Place it on a shaker and shake for about 10 minutes to seal.
4. Remove the sealed membrane and clean the protein membrane 2-3 times with washing solution, then proceed with Western Blot experiments such as dilution of primary antibody.
