概要
Triglycerides are saponified by KOH to yield glycerol and fatty acids. The glycerol is then oxidized by periodic acid to formaldehyde. In the presence of ammonium ions, formaldehyde condenses with acetylacetone to generate a yellow product, which exhibits a characteristic absorbance peak at 420 nm. The triglyceride content can be quantitatively determined by measuring the change in absorbance at this wavelength.
