Tissues containing bone or calcified foci are not suitable for direct paraffin embedding and sectioning, because the differing densities of calcium deposits and paraffin make it difficult to obtain intact sections. Calcium‑containing tissues should undergo decalcification after fixation, or fixation and decalcification may be carried out simultaneously, before proceeding to dehydration, clearing, infiltration, embedding, sectioning, and other downstream procedures. Commonly used decalcifying reagents include organic acids, inorganic acids, ethylenediaminetetraacetic acid (EDTA), and electrolytic decalcification methods. During decalcification, the degree of decalcification should be monitored periodically to avoid excessive decalcification, which may cause additional tissue damage and compromise subsequent staining results. This product (Decalcification Endpoint Detection Kit) consists of ammonium hydroxide, ammonium oxalate, and other components. The principle is as follows: after the addition of ammonium oxalate, if no precipitate forms, it indicates that calcium ions have been sufficiently removed, i.e., the decalcification endpoint has been reached. This reagent is intended for research use only and is not suitable for clinical diagnosis or other applications.
