Aladdin's Endogenous Peroxidase Blocking Buffer can be used to block endogenous peroxidase in cells or tissues in Immunohistochemistry (IHC), Immunocytochemistry (ICC) and in situ Hybridization (ISH) assays.Endogenous peroxidase is widely present in some cells and tissues including red blood cells, kidney and liver tissues, which results in a high background and even false positive results when samples are assayed using the peroxidase method. Therefore, it is advisable to to block endogenous peroxidase in cells or tissues with an appropriate peroxidase blocking buffer before staining to eliminate the interference from endogenous peroxidase. This product has been diligently optimized to provide good blocking efficacy, effectively reducing the staining background without diminishing the specific signal. It is better than conventional hydrogen peroxide solutions in blocking endogenous peroxidase in many cases (Figure 1).Figure 1. Effectiveness of Aladdin's Endogenous Peroxidase Blocking Buffer . HRP-labeled secondary antibodies were diluted at 1
Precautions:
If staining background is still high after using this product, Enhanced Endogenous Peroxidase Blocking Buffer can be attempted. For some experiments that are incompatible with peroxidase assay system, consider using an alkaline phosphatase assay system. This product is irritating. Please take proper protections when handling. This product is for R&D only. Not for drug, household, or other uses. For your safety and health, please wear a lab coat and disposable gloves during the operation.
Instructions for Use:
1. For Immunohistochemistry (IHC) and Immunocytochemistry (ICC)a. Wash samples for 3-5min with Immunostaining Wash Buffer or a liquid with cell membrane permeability.b. Discard the wash buffer, add an appropriate amount of Endogenous Peroxidase Blocking Buffer dropwise to cover samples evenly and completely, and incubate at room temperature for 5-10 min or at 37ºC for 2-5 min.c. Discard the Endogenous Peroxidase Blocking Buffer and wash samples 2-3 times for 1-3 minutes each with a wash buffer or other appropriate solutions such as PBS. Subsequent steps such as blocking and primary antibody incubation can then be performed.2. For in situ Hybridization (ISH) a. Before performing probe hybridization, add an appropriate amount of Endogenous Peroxidase Blocking Buffer dropwise to cover samples evenly and completely, and incubate at room temperature for 5-10 min or at 37ºC for 2-5 min.b. Discard the Endogenous Peroxidase Blocking Buffer and wash 2-3 times for 2-5 minutes each with the wash buffer for in situ Hybridization. Hybridization can be perform subsequently.
