Plant stomatal detection experiment by fixation method
Plant stomatal detection experiment by fixation method
Through the experiment, master the principle and method of observing the stomatal opening and closing condition of plant leaves by the fixation method.
Operation method
Plant stomatal detection experiment by fixation method
Principle
Anhydrous ethanol can make the plant cells quickly dehydrated and dead, thus the cell wall is hardened, the cell shape is fixed, and the stomata can be preserved in their original form, which is favorable for later microscopic research, and the plant material can also be preserved for a long time.
Materials and Instruments
Plant leaves Move 1. Tear off the epidermis of the leaf with tweezers (a small cut can be made with a scalpel to facilitate tearing), and quickly (the faster, the better) put it into anhydrous ethanol for fixation and preservation. Place the fixed epidermis on a slide, add a few drops of anhydrous ethanol, cover with a coverslip, and examine it microscopically. For more product details, please visit Aladdin Scientific website.

2. The key to the success of this method lies in the rapid action, the whole process (torn off and immersed in anhydrous ethanol) should not be more than 1-2 seconds, otherwise the cell wall of the defense cells can not be quickly dehydrated and hardened and difficult to maintain the original shape, at the same time, this method can only be used for epidermis is easy to tear the plant tissue.
