Proteins Involved in Apoptosis

Apoptosis is a regulated process of programmed cell death in which cells initiate internal signaling pathways that lead to their own elimination. This mechanism is essential for normal development and tissue homeostasis, but it can also be triggered by external stressors such as hypoxia, ionic disturbances, or infection.


Disruption of apoptosis contributes to a wide range of diseases, including tissue atrophy, neurodegenerative disorders, autoimmune conditions, and cancer. In many cancers, apoptotic pathways are suppressed, enabling cells to survive beyond their natural lifespan. This prolonged survival allows the accumulation and propagation of damaged or misfolded proteins during continued cell division.


Key proteins controlling apoptosis are frequently mutated or dysregulated in cancerous cells. Notable examples include the tumor suppressor p53 and anti-apoptotic regulators such as Bcl-2 and XIAP. p53 functions by halting the cell cycle at the G1 checkpoint following DNA damage, promoting either DNA repair or the initiation of apoptosis. Negative regulators like MDM2 and related proteins inhibit p53 activity, while blocking these regulators restores p53 function.


Members of the Bcl family (e.g., Bcl-2, Bcl-xL) act as apoptosis inhibitors by preventing the release of cytochrome C and suppressing caspase activation, both of which are central events in apoptotic signaling. Similarly, inhibitor of apoptosis proteins (IAPs) such as XIAP and survivin promote cell survival by directly binding caspases and blocking their activation.
