Trypan blue rejection assay

Summary

Taipan Blue is a cellular reactive dye that is commonly used to test the integrity of cell membranes. It is also used to detect the survival of cells. Live cells are not colored blue, while dead cells are colored light blue.

Principle

Taipan blue dye can not pass through the normal cell membrane of living cells, so living cells are not colored, but the cell membrane permeability of dead cells increases, allowing the dye to pass through the cell membrane into the cell, so that the dead cells are colored blue. It is the most commonly used method to detect cell viability.

1. Dilute the cells to be stained to the desired concentration. (The method and concentration range are the same as for cell counting.)
2. Add a small drop of freshly prepared stain solution for every 0.1 ml of cell suspension and stain for 3-5 min at room temperature.
3. Stained cellular material, take a drop of cell suspension and place it on a slide, cover the slide and place it under a high magnification microscope.
4. Dead cells are light blue and enlarged and have no luster; living cells are not colored and retain their normal shape and luster. Count 100-200 white blood cells.
5. Count unstained cells. Cell viability can be calculated according to the formula. Cell viability (%) = number of unstained cells/total number of cells observed × 100.

Caveat

1. the staining time should not be too long, otherwise the living cells will also gradually accumulate the dye and become stained, making the test result low.

2. Cell counting and viability testing can also be combined with cell counting methods.
