Panoramica

Amylase (AMS), also known as 1,4-α-D-glucan hydrolase, is a general term for a class of enzymes capable of catalyzing the hydrolysis of starch and glycogen. Based on the substrate used for detection, its assay methods are primarily divided into two categories: one employs natural starch as the substrate (such as the iodine-starch method adopted by this kit), and the other utilizes defined substrates (such as maltopentaose or 4-nitrophenyl maltoside (4-NP-G)). This kit is based on the classic iodine-starch colorimetric principle. The α-amylase in the sample catalyzes the hydrolysis of the α-1,4-glycosidic bonds in starch molecules, producing glucose, maltose, dextrins, and other products. In the reaction system, iodine solution binds to the unhydrolyzed starch to form a blue complex. The higher the α-amylase activity, the less starch remains, resulting in a lighter blue color. By measuring the absorbance at 660 nm using a microplate reader and comparing it with a blank control, the amylase activity units in the sample can be calculated. It is suitable for detecting endogenous amylase activity in various biological samples such as cell or tissue lysates/homogenates, plasma, serum, and urine. This product is intended for research use only and is not suitable for clinical diagnosis or other purposes.
