Plant tissue culture, also known as axenic culture or in‑vitro culture technology, is based on the theory of plant cell totipotency. Using explants including isolated plant organs (roots, stems, leaves, flowers, shoot‑tips, fruits, etc.), tissues (cambium, epidermis, cortical‑pith cells, endosperm, etc.), cells (megaspores, microspores, somatic cells, etc.) and protoplasts, this technique enables the induction of callus, adventitious buds and adventitious roots under suitable artificial culture medium and environmental conditions, and finally regenerates intact plantlets.
B5 medium is characterized by low ammonium‑salt content and relatively high nitrate and thiamine levels. It is mainly applied to the cultivation of dicotyledons, especially woody plants. B5 Vitamin Solution (1000×) is mainly composed of myo‑inositol, nicotinic acid, pyridoxine hydrochloride, thiamine and other components, and serves as a common medium supplement. This reagent is intended for research‑only use and shall not be used for clinical diagnosis or other purposes.
1. Directions for use: Add 1 mL of this product to 1 L of medium solution and mix thoroughly. Adjust the pH to the required value, generally 5.8±0.2.
2. If necessary, the medium can be autoclaved for plant‑tissue‑culture applications. Additional carbon sources, gelling agents, plant hormones or plant‑growth‑regulators may also be supplemented.
Precautions:
1. This product is used as an additive for corresponding plant‑tissue‑culture basal salt medium.
2. Select a suitable culture medium according to different experimental materials and plant species; optimal conditions are best determined experimentally.
3. Use the reagent as soon as possible after opening to avoid impairing subsequent experimental results.
4. If strict sterility is required, autoclave again at 121 ℃ for 15‑20 min.
5. For your safety and health, wear a lab coat and disposable gloves during operation.
