Brefeldin A (BFA) is an intracellular protein transport inhibitor. Adding BFA during in vitro cell activation blocks protein transport to the Golgi Complex, causing accumulation in the Endoplasmic Reticulum, thereby enhancing the detection signal for intracellular cytokine staining. This method is particularly useful and effective for staining most cytokines in mouse and human cells. Optimal conditions, including the duration of treatment, are cell type and assay-dependent. Typically, BFA is added for the last few hours up to 4-24 hours prior to cell harvest. Researchers are advised to evaluate the performance of BFA, along with other transport inhibitors like monensin, in their specific assay system. Brefeldin A Solution is commonly supplied as a 1,000X concentrate and should be diluted to a 1X working concentration in cell culture medium.
