Bromelain, often referred to simply as bromelain, is a proteolytic enzyme found in pineapple plants. It can be extracted from the fruit stem, leaves, and peel of pineapples, and is obtained through processes such as purification, concentration, enzyme immobilization, and freeze-drying. Bromelain is widely used in the food, pharmaceutical, and other industries. By using different coupling methods such as adsorption, ionic bonds, and covalent bonds, the enzyme can be linked to an insoluble carrier to prepare immobilized enzymes, which effectively prevents enzyme inactivation and facilitates storage and transport. The detection principle of this kit is based on the hydrolysis of the substrate casein catalyzed by bromelain, producing tyrosine. Tyrosine contains a phenolic hydroxyl group that can react with Folin reagent in the Folin reaction. Specifically, under alkaline conditions, Folin reagent is readily reduced quantitatively by phenolic compounds, producing a mixture of tungsten blue and molybdenum blue, which exhibits varying shades of blue. The absorbance is measured at 680 nm. The magnitude of the absorbance is related to the amount of tyrosine; a higher absorbance indicates a higher tyrosine content, meaning more casein has been broken down by bromelain, reflecting higher enzyme activity. This kit is primarily used for detecting the activity of bromelain in plant tissues, especially in pineapple homogenates.
