Protein denaturation and sample preparation for enzymatic digestion are the most critical experimental steps in peptide mapping, glycan analysis of biopharmaceutical antibodies, and proteomics research. Native proteins exist in a three-dimensional structure. To expose digestion sites within the higher-order structure and enable efficient interaction with proteases, proteins must be fully denatured, reduced, alkylated, and subjected to necessary enzymatic reactions. Traditional denaturants such as urea, guanidine hydrochloride, or SDS can cause certain chemical modifications to proteins after denaturation, such as carbamylation, leading to interference and errors in polypeptide identification. The use of BT Surfactant avoids such modification issues. Reconstitution: Reconstitute in pH 8.0 buffer or ultrapure water. Add 500 μL buffer to 5 mg BT Surfactant to obtain a 1% solution. pH Range: pH 7–9 for protein denaturation or enzymatic reactions. BT Surfactant degrades and precipitates in solution at pH 2–4. Recommended Usage: Add 1–2% BT Surfactant to samples for protein denaturation. Perform proteolytic digestion in 0.1% BT solution after denaturation. Effect of BT Surfactant Concentration on Protease Activity: BT concentration <2%: no effect on trypsin activity. BT concentration <1%: no effect on Lys-C activity. BT concentration <1%: no effect on rPNGase F glycosidase activity. Effect of Temperature: Incubation at 37 °C for 4 h: BT degradation peak <1% in MS. Incubation at 95 °C for 10 min: BT degradation peak <5% in MS.
