Carbonate Fuchsin Staining Solution differs from modified carbol fuchsin staining solution. Composed mainly of carbolic acid and basic fuchsin, carbol fuchsin staining solution is primarily applied to sputum smear or swab staining of Mycobacterium tuberculosis and Gram staining, and rarely used alone. Also known as Carbol fuchsin, modified carbol fuchsin staining solution consists of phenol, basic fuchsin, sorbitol and oxidants, commonly used for cell nucleus staining as well as fixation and staining of chromosomes.
Carbol fuchsin staining solution is mainly used for acid-fast bacilli staining and Gram staining, and can also be used independently.
Instructions for Use:
1. Acid-fast Staining:
① Collect the test sample with an inoculating loop, spread it evenly on a glass slide and air-dry naturally.
② Add carbol fuchsin staining solution and stain for 5 to 10 minutes without heating, then rinse with distilled water.
③ Decolorize with decolorizing solution until no red color remains, followed by distilled water rinsing.
④ Stain with methylene blue staining solution for 30 to 60 seconds and rinse with distilled water.
⑤ Blot moisture gently and air-dry the specimen.
⑥ Examine under an oil immersion objective.
2. Paraffin Section Staining:
① Deparaffinize paraffin sections and rehydrate with water.
② Apply carbol fuchsin staining solution and stain for 3 to 5 minutes, then rinse briefly with running water.
③ Differentiate with hydrochloric acid ethanol for 5 to 10 seconds and rinse slightly with running water.
④ Clear with xylene directly and mount with neutral balsam.
Precautions:
1. Microscopic observation shall be conducted immediately after staining.
2. Wear lab coat and disposable gloves during operation for personal safety and health protection.
