Recombinant Carboxypeptidase B is a proteolytic enzyme that specifically cleaves the N‑terminus of basic amino acids (Lysine, K and Arginine, R) at the C‑terminus of proteins. It has a molecular weight of 34.7 kDa and an optimum pH of 7.5‑9.0. Its activity is competitively inhibited by arginine and lysine, and is also inhibited by metal‑ion chelators such as EDTA.
The amino‑acid sequence of this product is identical to that of porcine pancreatic‑derived carboxypeptidase B, and it exhibits identical enzymatic properties to animal‑derived carboxypeptidase B.
Expressed in the low‑endotoxin *E. coli* CleanTAK™ strain, recombinant Carboxypeptidase B features extremely low endotoxin levels and is free of any animal‑derived pathogens. It has a broad scope of applications, especially for research, insulin manufacturing, medical diagnostics and other fields.
Instructions for Use
Solution Preparation: Reconstitute recombinant Carboxypeptidase B in sterile water or 25 mM Tris‑HCl (pH 7.6) to a concentration of 1‑10 mg/mL.
Enzyme Dosage: Enzyme : target protein (mass ratio) = 1:50‑1:1000; optimum pH: 7.5‑9.0.
Solution Storage: The prepared solution may be used on the day of preparation or stored at 2‑8 °C. For long‑term storage: aliquot the reconstituted recombinant Carboxypeptidase B stock solution and store at −20 °C or lower. Avoid repeated freeze‑thaw cycles.
Note: This product is supplied as lyophilized powder. Centrifuge the vial prior to opening to collect all powder at the bottom.
Product Applications
Possessing enzymatic properties identical to animal‑derived carboxypeptidase B, it can be applied in various biotechnological processes:
‑ Production of recombinant insulin and its analogues;
‑ Determination of C‑terminal amino acids of proteins;
‑ Manufacturing of other recombinant polypeptides;
‑ Enzymatic synthesis of certain special compounds.
Product Advantages
‑ Extremely low endotoxin: Recombinant Carboxypeptidase B expressed by low‑endotoxin *E. coli* CleanTAK™ strain has extremely low endotoxin content.
‑ Animal‑free: Recombinant enzyme without any animal‑derived raw materials.
‑ Consistent quality: Mass production with negligible batch‑to‑batch variation.
‑ High purity: Residuals of microorganisms, host‑cell protein, exogenous DNA and metal ions are below the limits for biological products.
Precautions
‑ Enzyme activity is subject to competitive inhibition by arginine and lysine, and inhibition by metal‑ion chelators (e.g. EDTA).
‑ For personal safety, wear a lab coat and gloves during handling.
‑ For research and manufacturing use only. Not for human administration or any other purposes.
