2. Principles and Uses
Trypticase (pancreatic digest of casein), peptic digest of meat, and pancreatic digest of heart provide nitrogen sources, growth factors, minerals, amino acids, etc. Yeast extract powder and corn starch supply vitamins, especially B‑group vitamins essential for bacterial growth. Sodium chloride maintains osmotic equilibrium. Agar is the solidifying agent.

4. Preparation Method
Weigh 44.0 g of the medium and transfer to 1 L of purified water. Heat to boiling with frequent agitation until completely dissolved. Dispense into suitable containers and autoclave at 121°C for 15 min. Cool to 45–50°C. Under aseptic conditions, add 1 mL of gentamicin sulfate solution (KS0005, 2 mg/mL) per 100 mL of medium, mix well, and pour into Petri dishes for use.
