Staining methods for the detection of Cryptosporidium oocysts include the auramine-phenol staining method and the modified acid-fast staining method, among others. The Auramine-Phenol Staining Solution is composed of auramine and phenol, and is suitable for staining Cryptosporidium oocysts in fresh fecal specimens or specimens fixed with formalin. This reagent is for research use only and is not suitable for clinical diagnosis or other purposes.

Operating Steps (For Reference Only)
Prepare a thin fecal smear on a glass slide and allow it to air dry. Fix with methanol for 2–3 minutes.
Apply Auramine-Phenol Staining Solution onto the air-dried fecal film and let stand for 10–15 minutes. Rinse with water.
Apply Differentiation Solution onto the fecal film and let stand for 1 minute. Rinse with water.
Apply Oxidizing Solution onto the fecal film and let stand for 1 minute. Rinse with water.
Allow the slide to dry and examine under a fluorescence microscope.
Staining Results
Under low magnification fluorescence microscopy, oocysts appear as small, round bright spots emitting a milky white fluorescence.
Under high magnification fluorescence microscopy, oocysts appear milky white or slightly greenish. The oocyst wall is thin, and most oocysts show a deeply stained periphery with a paler center, giving a ring‑like appearance.
Precautions
Some specimens may show non‑specific fluorescent particles; careful differentiation is required.
