Di‑4‑ANEPPDHQ is a dye that rapidly responds to changes in membrane potential. It has no intrinsic fluorescence and emits fluorescence only upon binding to cell membranes. It is sufficiently fast to detect transient (millisecond‑scale) membrane‑potential changes in excitable cells. The magnitude of fluorescence change of Di‑4‑ANEPPDHQ is relatively consistent across various tissues, cells and membrane models, typically exhibiting a 10 % fluorescence shift per 100 mV change.
Procedure
1. Preparation of DMSO stock solution: Prepare 1‑10 mM stock solution with DMSO. Note: Aliquot unused stock solution and store at −20 °C; avoid repeated freeze‑thaw cycles.
2. Preparation of working solution: Dilute the stock solution with an appropriate buffer (e.g., serum‑free medium, PBS) to obtain a working solution of 10‑15 μM. Note: The final working concentration should be optimized according to individual experimental systems. It is recommended to explore the optimal concentration within a 10‑fold range of the suggested concentration.

1. Briefly centrifuge the vial to spin down contents to the bottom of the tube before use.
2. Fluorescent dyes are susceptible to photobleaching. Minimize light exposure to slow down photobleaching.
