Overview

DiIC16(3) is a long-chain carbocyanine dye. Carbocyanine dyes are widely used as lipophilic tracers for labeling cells, organelles, liposomes, viruses, and lipoproteins. Long-chain carbocyanines include DiO (DiOC18(3)), DiI (DiIC18(3)), DiD (DiIC18(5)), and DiR, as well as the dialkylaminostyryl dye DiA (4-Di-16-ASP), which are used to label membranes and other hydrophobic structures. The alkyl substituent of DiIC16(3) (C16) is shorter than that of DiI (C18). They exhibit extremely high extinction coefficients, environment-dependent fluorescence, and short excited-state lifetimes in lipid environments. They are weakly fluorescent in water but highly fluorescent and reasonably photostable when bound to membranes or lipophilic biomolecules. These optical properties make them ideal for plasma membrane staining. Once applied to cells, these dyes diffuse laterally within the plasma membrane, resulting in staining of the entire cell.

DiO, DiI, DiD, and DiR exhibit distinct green, orange, red, and infrared fluorescence, respectively, facilitating multicolor imaging and flow cytometry analysis of live cells. DiO and DiI can be used in standard FITC and TRITC fluorescence channels, respectively. Among them, DiI and its derivatives are the most commonly used because they generally show very low cytotoxicity. In addition, DiI is widely used for the assay of lipoproteins such as LDL and HDL. The lipophilic aminostyryl dye DiA is also commonly used for neuronal tracing.
