Product Introduction:
The EDTA antigen repair solution (50x) uses widely used EDTA, which has the ability to repair protein crosslinks caused by fixatives such as paraformaldehyde. Whether it is paraffin sections, frozen sections, or cell slides, as long as the sample is fixed with paraformaldehyde, formaldehyde, or other aldehyde reagents, antigen repair will effectively remove protein crosslinks, fully expose antigen epitopes, and greatly improve immunostaining efficiency.
Operation process:
1. Before preparing the reagent for use, it is necessary to dilute EDTA antigen retrieval solution (50 x) with distilled water or ultrapure water to prepare an antigen retrieval solution (1 x).
2. Antigen repair
2.1 For paraffin sections:
2.1.1 Dewaxing: The slices are dewaxed in xylene for 2 × 5 minutes. Anhydrous ethanol for 2 × 5 minutes. 90% ethanol for 2 × 5 minutes, 70% ethanol for 5 minutes. Distilled water for 2 × 5 minutes.
2.1.2 Antigen retrieval: Soak the slices in antigen retrieval solution (1 ×) and heat them at 95-100 ° C for about 10-30 minutes (the optimal heating time needs to be explored according to different samples and target proteins).
Note: The antigen repair solution (1 ×) needs to be preheated to 95-100 ° C before use. Heating can be done using a water bath or microwave. If using a microwave oven for heating, be careful to avoid boiling the repair solution. Then cool to room temperature for approximately 20-30 minutes. Wash 1-2 times with immunostaining detergent, each time for 3-5 minutes. Subsequent immunostaining steps such as sealing can be performed.
2.2 Frozen sections: Wash the sections with immunostaining detergent for 5 minutes. The process of antigen repair is similar to the antigen repair process of paraffin sections.
2.3 For antigen repair of other samples, the steps for paraffin sectioning can be followed.
