Panoramica
MS-grade Glu-C protease specifically cleaves peptide bonds at the C-terminus of glutamic acid (E) or aspartic acid (D) residues. This enzyme is suitable for in-solution digestion and is not recommended for in-gel digestion. It exhibits higher specificity for glutamic acid residues in ammonium bicarbonate or ammonium acetate buffers, while it can cleave both glutamic acid and aspartic acid residues in phosphate buffer.
Reconstitution Solution:
Reconstitute Glu-C protease with ultrapure water.
pH Range:
rGlu-C exhibits optimal activity at pH 4.0–9.0.
Application:
rGlu-C protease specifically cleaves aspartic acid (D) and glutamic acid (E) residues in denatured proteins.
In-Solution Digestion SOP:
Use 50 μg of Glu-C protease at a protein-to-protease ratio (w/w) of 25:1. Incubate overnight at 37°C for 13–16 hours at pH 8.5.
