Alginate encapsulation experiments

Summary

Alginate is one of the causes of SS, and an animal model similar to human SS was induced by alginate. Methods: Sixteen New Zealand white rabbits immunized with alginate were randomly divided into two groups, A1 and B1, and six normal New Zealand white rabbits in each of the groups A2 and B2 served as the control group, and were injected with alginate through the parotid ducts at intervals of 1d, and were tested for similarity to human SS in terms of the amount of saliva, the Schirmer test, the subpopulation of T lymphocytes, parotid imaging, and histopathology, etc. After 2w (Group A) and 4w (Group B), the results showed that human SS was similar to human SS in terms of changes in salivary volume, T lymphocytes and histopathology. subpopulations, parotid imaging and histopathologic changes.

Move

1. Dissolve sodium alginate in sterile saline to a final concentration of 1.5%;2. Collect the cultured tumor cells, wash them once with serum-free medium, and resuspend the cell precipitate in 1.5% sodium alginate solution;3. Slowly drop the above tumor cell suspension into a magnetically stirred 250 mM CaCl2 solution using a 1 ml sampling gun to form milky white alginate beads. Continue to let it stand in 250 mM CaCl2 for 30 min and it is ready for use. The above steps were performed under sterile conditions;4. 7 days after the 4th immunization, mice were anesthetized by intraperitoneal injection of 0.1 ml of sodium phenobarbital (100 mg/kg). After anesthesia, the mice were placed on a dissection table, the skin on the back was cut open, and 4 alginate-encapsulated pellets were implanted subcutaneously, the skin was closed, and surgical adhesive film was applied externally;5. Days later, mice were injected with 100 μl (100 mg/kg) of FITC-labeled dextran (FITCDextran) via the tail vein;6. 20 min later, the animals were executed, the alginate pellet was taken out, 1 ml of saline was added at room temperature, the pellet was cut and ground, 1 ml of saline was added, the specimen was mixed well, left for 1 h, and centrifuged at 1500 rpm for 5 min. The supernatant was taken and measured by fluorescence enzyme marker;7. Standard curves were prepared with different concentrations of FITC-Dextra n.
