1. Staining: acetaldehyde red
Naked eye observation:
This specimen has different shades of color and many darker stained filaments can be seen.
2. Low magnification observation:
The fibers are crossed into a network, and the cells are scattered between the fibers. Select the lightly colored parts and turn to high magnification observation.
3. High magnification observation:
Note the distinction between two types of fibers and two types of cells.
(1) Collagen fibers: numerous, stained pink. The fibers are thick, branched, and wavy in the natural relaxed state.
(2) Elastic fibers: small in number, thin and straight, also with branches, stained darker purple-blue, refractive, elastic fibers straight, curled at the broken end.
(3) Fibroblasts: large cells with multiple sharp protrusions and unclear cell margins. The cytoplasm is weakly basophilic, the nucleus is large and ovoid, and the staining is light.
(4) Macrophages: the cell shape is variable, round, oval or irregular, the border is clearer, some cells can be seen pseudopods. The cytoplasm is eosinophilic and contains blue placental blue particles and vacuoles of different sizes. The nuclei are more parabolic, smaller and darker in color.
(5) Stroma: The spaces between fibers and cells are filled with stroma (dissolved).
