Principle

Ferrous heme in hemoglobin has peroxide E-like activity. It can catalyze the release of neo O2 from H2O2 to oxidize the receptor o-toluidine to o-toluazole and show blue color.

Materials and Instruments

o-Toluidine H2O2

Move

1、Pick a small amount of feces with a bamboo skewer and apply it on a sterilized cotton rod or white porcelain plate.

2、Drop 0.15g/L o-toluidine ice acetic acid solution 2-3 drops on the stool.

4、Observe the results within 2 minutes blue-brown for positive

Caveat

l. Fecal specimens should be examined in a timely manner to avoid a decrease in sensitivity.

2、3%H2O2must be effective, positive control should be carried out.

3， 3 days before the experiment, fasting animal blood, meat, liver, etc. to avoid false positives.

4， the apparatus must be heat-treated to avoid contamination of H2O2enzyme.

Common Problems

Experimental judgment results:

(i) No color development after 2 minutes

(+) change from light blue to blue after 10s

(+ +) light blue-brown to blue-brown after adding reagent

(+ + + +) immediately blue-black brown

(＋＋＋＋) immediately blue-blackish brown
