The basic procedure for serum and plasma preparation experiments can be divided into the following steps:
(a) Serum isolation A
Whole blood obtained from the retroocular vein or abdominal a
orta.
B After standing in EP tubes at room temperature for more than 1 hour or in a 37 ℃ water bath for about 1 hour.
C Refrigerate at 4°C for 2 hours or overnight.
D Centrifuge at 3000 r/min for 10 minutes, the supernatant is serum, which can be stored at -80 ℃ after separation to avoid repeated freezing and thawing.
(ii) Plasma separation
A Plasma is the upper clear liquid containing fibrinogen and other coagulation components obtained by centrifugation of anticoagulated whole blood. Since plasma reflects the original state of blood components and the yield is larger than that of serum, it is widely used in the component testing of blood. Using dried heparin tubes or anticoagulant tubes containing a small amount of liquid heparin, whole blood obtained from the posterior ophthalmic vein or the abdominal aorta is mixed with anticoagulant by spinning thoroughly (avoiding strenuous movements), and then centrifuged at 3000 r/min for 10 minutes to obtain supernatant, and then centrifuged at 5000 r/min for 3 minutes, which gives a higher-quality plasma sample.
Caveat
1 Isolated plasma can be stored at 4 °C for several days and at -20 °C or -80 °C for longer periods. To determine lactate dehydrogenase activity, blood must be collected and measured immediately to obtain valid results.
