GBP6 Mouse mAb (Ab191420) - IHC
IHC analysis of esophagus tissue by GBP6 Mouse mAb (Ab191420).
IHC was performed using sections of the formalin-fixed paraffin-embedded esophagus tissue. Antigen was retrieved through addition of boiling Tris/EDTA buffer pH 9.0 in a pressure cooker for 3 min. Endogenous peroxidase activity was quenched by incubating the sections with 3% H2O2 for 30 min at room temperature. The sections were then incubated with GBP6 Mouse mAb (Ab191420) at 10 µg/mL at room temperature for 1 h. Poly-peroxidase conjugated goat anti-mouse IgG was used as the secondary antibody. Diaminobenzidine was used as the chromogen. The section was counterstained with hematoxylin. A tissue section incubated with phosphate-buffered saline followed by incubation with the secondary antibody was used as the background control.
Result: Squamous epithelial cells are positively stained at the cytoplasm.

Application Protocols

Item-specific tested applications, recommended dilutions, antigen retrieval, incubation times, and positive control samples: Not specified for this item; refer to CoA/Spec Sheet.

General framework (for user planning only; not product claims):

  • Western blot: Prepare lysates, denature/reduce as appropriate, transfer to membrane, block, incubate with primary anti-GBP6 mAb, wash, incubate with anti-mouse secondary, develop. Titrate antibody and optimize blocking/washing.
  • Immunoprecipitation: Couple antibody to Protein G/A resin or use crosslinking to reduce heavy/light chain contamination; incubate with clarified lysate, wash, elute under native or denaturing conditions.
  • IF/ICC: Fix cells (e.g., PFA), permeabilize if intracellular target, block, incubate with primary, wash, fluorescent secondary, mount; adjust fixation/permeabilization for GBP6 epitope.
  • IHC: Perform antigen retrieval as needed, block endogenous activity, incubate with primary, detect via HRP/AP or fluorescence; include isotype and no-primary controls.

Please verify species reactivity and optimal conditions empirically and consult the CoA/Spec Sheet for any product-specific recommendations.

Biological Roles

Target context (literature; not product performance claims):

  • GBP6 (Guanylate-Binding Protein 6) is a member of the dynamin-like large GTPase family induced by interferons, particularly IFN-γ. GBPs participate in cell-autonomous immunity, membrane remodeling, and responses to intracellular pathogens in various systems.
  • Structural/biochemical features: GBPs bind and hydrolyze GTP, can dimerize/oligomerize, and may associate with cellular membranes via lipid modifications or specific domains (details vary by GBP family member; exact features of GBP6 depend on isoform and species).
  • Cellular localization: GBPs often localize to cytosol and can be recruited to pathogen-containing vacuoles or membranes upon activation (literature; GBP6-specific localization patterns should be confirmed experimentally).
  • Biological processes: interferon signaling, innate immune effector functions, and regulation of inflammatory pathways have been associated with GBPs broadly; GBP6-specific roles are an active area of research.

Research implications:

  • An anti-GBP6 monoclonal antibody enables interrogation of GBP6 expression dynamics following cytokine stimulation, mapping of subcellular localization, and discovery of interacting partners by IP/Co-IP and MS.

Note: The above summarizes general scientific knowledge about GBP6/GBP family proteins and does not imply validated application performance for this item. Always verify species reactivity and assay suitability empirically.

Buffer Applications

This product is an antibody rather than a small-molecule buffer component. Nevertheless, buffer choice is critical for antibody performance.

General guidance (not item-specific):

  • Storage/working buffers: Phosphate-buffered saline (PBS, pH ~7.2–7.6) or Tris-buffered saline (TBS, pH ~7.4–8.0) are common. Additives such as 0.01–0.05% non-ionic surfactant can reduce nonspecific adsorption; presence/absence for this item is not specified.
  • Blocking buffers for immunoassays: 1–5% BSA, non-fat milk, or serum can reduce background; avoid blockers that share epitopes with targets or interfere with detection.
  • For conjugation workflows: carrier-free antibodies are often buffer-exchanged into additive-free PBS or bicarbonate, or specialized buffers compatible with NHS/SMCC chemistries; ensure removal of amines/thiols as required by the chemistry.
  • pH considerations: most IgGs retain activity near neutral pH; extreme pH may denature or alter binding.

Item-specific buffer composition, pH, and any stabilizers/preservatives: Not specified for this item; refer to CoA/Spec Sheet.

Green Alternatives

Antibody products do not typically involve volatile organic solvents, but sustainability considerations still apply.

Greener considerations (general):

  • Carrier-free formulation: avoids additional proteins and biocides that may complicate waste handling. It also reduces additive-derived interferences, enabling lower reagent use through efficient conjugation.
  • Preservative-free/azide-free handling: minimizes disposal of toxic azide; however, aseptic technique and aliquoting are needed to mitigate contamination risk (item-specific preservative status is not specified).
  • Concentrated stocks: using higher-concentration antibody stocks can reduce packaging and cold-chain volume (concentration not specified for this item).
  • Cold-chain optimization: consolidate shipments and use reusable cold packs where institutional policy allows; this product ships on dry ice + cold packs as specified.

Comparison (general) — environmental and workflow trade-offs:

  • Carrier-free mAb vs azide/BSA-stabilized mAb:
    • Environmental: reduced hazardous preservative load vs increased need for sterile handling.
    • Workflow: improved conjugation and assay specificity vs potentially lower long-term bench stability without carriers.

Institutional tips:

  • Plan experiments to minimize freeze–thaw cycles and waste.
  • Use validated recycling/return programs for cold-chain materials when available.
Pharmaceutical Uses

This product is supplied strictly for research use only; it is not intended for diagnostic, therapeutic, or human/animal administration.

Context for formulation science (general; not item-specific):

  • Monoclonal antibodies can serve as analytical reagents and process controls in biopharmaceutical development (e.g., assay standards, positive controls), provided they meet internal quality requirements. This product is not supplied as GMP-grade.
  • Carrier-free status is advantageous for custom conjugation to enzymes or fluorophores used in in-process analytical assays or platform methods.
  • Regulatory note: For any use beyond basic research (e.g., QC of clinical materials), institutions typically require qualification/validation and supplier documentation beyond standard research-grade COA/SDS.

Item-specific pharmacopeial status, excipients, endotoxin/bioburden limits: Not specified for this item; refer to CoA/Spec Sheet.

Physical Properties

Item-specific physico-chemical specifications (formulation, concentration, excipients, pH, protein content, UV cutoff, absorptivity): Not specified for this item; refer to CoA/Spec Sheet.

General/literature properties for monoclonal IgG (for context only; not item specifications):

  • Molecular weight: ~150 kDa (intact IgG); fragmentation yields ~50 kDa heavy chain and ~25 kDa light chain under reducing SDS-PAGE (literature).
  • Isoelectric point (pI): commonly ~6–9 depending on subclass and sequence (literature; item-specific pI not specified).
  • Solubility: typically highly soluble in physiological buffers (e.g., PBS, TBS) at mg/mL levels; salting-out at high ammonium sulfate (literature).
  • UV absorbance: A280 1.0 ≈ 0.7–1.5 mg/mL for IgG depending on sequence; ε280 ~ 200–230 kDa−1·cm−1 (literature; item-specific value not specified).
  • Stability: generally stable 4–8 °C short term; frozen at −20 to −80 °C for extended storage; avoid repeated freeze–thaw (literature; see Storage tab for item-specific guidance).

Physical appearance: Not specified for this item.

Note: Do not interpret the above as product specifications; consult the CoA/SDS for definitive item data.

Quality and Grades

Item-specific quality notes from Product Data:

  • Grade/Purity: Carrier Free, ExactAb™, Validated.

What these designations generally imply (context; not adding unstated specs):

  • Carrier Free: typically indicates the antibody is provided without carrier proteins (e.g., BSA) or other additives that can interfere with conjugation, functional assays, or MS-based workflows. This is advantageous for custom labeling (enzymes, fluorophores) and for applications sensitive to protein blockers. Exact excipients and buffer composition are not specified for this item; refer to CoA/Spec Sheet.
  • ExactAb™: denotes a controlled, standardized monoclonal format intended to deliver lot-to-lot consistency in affinity and specificity. This supports reproducibility across studies.
  • Validated: indicates the antibody has undergone internal qualification by the manufacturer; specific validated applications, species reactivity, and performance metrics are not listed here and should be confirmed on the CoA/Spec Sheet.

Batch-to-batch consistency:

  • Monoclonal antibodies inherently support consistent epitope recognition. ExactAb™ programs typically further control production parameters to reduce variability.

Documentation:

  • For chromatography traces, purity assessment (SDS-PAGE/SEC), endotoxin/bioburden, exact concentration, and any preservative status: Not specified for this item; refer to CoA/Spec Sheet.
Reaction and Applications

This product is a monoclonal antibody intended for research applications targeting GBP6. While the Manufacturer Applications field does not list specific tested uses for this SKU, typical research applications for a mouse anti-GBP6 mAb include (general guidance; verify suitability experimentally):

  • Immunoblotting (WB): detection of GBP6 in lysates under reducing or non-reducing conditions, depending on epitope accessibility. Optimize transfer conditions for high-MW GTPase family members; include appropriate positive controls (cells/tissues expressing GBP6) and negative controls (GBP6-knockdown/knockout).
  • Immunoprecipitation (IP)/Co-IP: enrichment of GBP6-containing complexes for downstream MS or western detection; use gentle, non-denaturing lysis where complex integrity is critical.
  • Immunofluorescence/Immunocytochemistry (IF/ICC): cellular localization studies; fixation and permeabilization conditions (e.g., PFA + Triton X-100) must be tuned to preserve GBP6 epitopes.
  • Immunohistochemistry (IHC): tissue distribution analysis; antigen retrieval conditions require optimization.
  • ELISA/Sandwich assays: as capture or detection reagent if epitope compatibility allows; carrier-free status is advantageous for direct conjugation to enzymes/fluorophores.

Controls and validation (general best practices):

  • Include isotype controls and secondary-only controls to judge specificity/background.
  • Confirm band size and antigen dependence using orthogonal methods (siRNA/CRISPR, overexpression, peptide block if available).

Note: Specific validated applications and conditions for this SKU are not provided; consult the CoA/Spec Sheet and perform in-lab optimization.

Reaction Conditions

Conventional chemical reaction conditions are not applicable to antibody use. For biochemical applications, general assay condition considerations include:

  • Temperature: Perform binding and washing steps typically at 4–25 °C depending on assay. Lower temperatures can reduce proteolysis and nonspecific binding; room temperature can speed kinetics. Item-specific recommendations are not provided.
  • pH and ionic strength: Near-neutral pH and physiological ionic strength (e.g., 0.15 M NaCl) help maintain antibody structure and reduce nonspecific interactions.
  • Detergents: Mild non-ionic detergents may be used in IF/ICC and IP lysis buffers to preserve complexes while limiting background. Optimize empirically based on GBP6 localization and membrane association.
  • Incubation times: Optimize for each assay and sample type; not specified for this item.
  • Antibody amount: Titrate to determine the minimum amount that yields specific, high S/N signal; not specified for this item.

Note: No item-specific, validated assay conditions are provided in the Product Data. Consult the CoA/Spec Sheet and optimize experimentally.

Safety and Handling

GHS/SDS information specific to this item:

  • Signal word, hazard statements, GHS classification, pictograms: Not specified for this item; refer to SDS for authoritative safety details.

General laboratory guidance for protein antibodies (good practice; not a substitute for SDS):

  • PPE: lab coat, gloves, and safety glasses to prevent exposure to preservatives, buffers, or microbial contaminants.
  • Handling: work aseptically to limit contamination. Avoid generating aerosols. If vial is lyophilized, reconstitute slowly to minimize foaming/denaturation.
  • Incompatibilities: avoid strong oxidants, proteases, and extreme pH (<4 or >10) that can denature proteins (general guidance).
  • Freeze–thaw: repeated cycles can cause aggregation and loss of binding; aliquot upon first thaw.
  • Spills: absorb with inert material; clean with suitable disinfectant (e.g., 70% ethanol), then water.
  • First aid (general):
    • Skin/eye contact: rinse with water for several minutes; remove contaminated clothing; seek medical advice if irritation persists.
    • Ingestion/inhalation: rinse mouth, move to fresh air; seek medical attention as needed.

Waste disposal: Follow institutional biosafety procedures for nonhazardous biological reagents; decontaminate containers if required. Always consult the product SDS for definitive hazard and disposal information.

Solvent Selection

For antibodies, the relevant “solvent” is the aqueous buffer system rather than organic solvents.

  • Polarity/miscibility: Antibodies are water-soluble macromolecules and are typically formulated in isotonic, buffered saline solutions. Miscibility in organic solvents is limited; high organic content can denature proteins.
  • Typical buffer systems (general guidance):
    • PBS or TBS at near-neutral pH (≈7.2–7.6) to maintain structure.
    • Low concentrations of non-ionic surfactants (e.g., polysorbate) are sometimes used to reduce aggregation; presence/absence not specified for this item.
    • For carrier-free preparations, absence of BSA/glycerol/azide facilitates downstream conjugations and cell-based assays sensitive to additives.
  • When to choose aqueous vs specialized matrices:
    • For conjugation: use additive-free buffers compatible with the chemistry (amine- or thiol-reactive). Desalt or exchange if needed.
    • For cell-based assays: avoid sodium azide and high osmolarity; ensure endotoxin is within acceptable limits (item-specific data not provided here).

Comparison (general):

  • Carrier-free mAb vs BSA-containing mAb: carrier-free minimizes background in ELISA and enables clean labeling; BSA-containing formats can enhance stability but may interfere with conjugation and MS.

Item-specific buffer composition and pH: Not specified for this item; refer to CoA/Spec Sheet.

Storage and Reconstitution

Item-specific storage and shipping:

  • Storage conditions: Store at −80 °C. Avoid repeated freezing and thawing.
  • Shipped in: Dry ice packs + Cold packs

Aliquoting and handling (general best practices; not item-specific requirements):

  • Upon receipt, spin down contents to collect material in the vial. If frozen, thaw gently on ice.
  • Immediately prepare single-use aliquots in sterile, low-protein-binding tubes to prevent loss and freeze–thaw damage. Refreeze aliquots at −80 °C promptly.
  • To minimize adsorption, keep protein concentrations reasonably high during storage; add inert protein only if compatible with downstream assays (this product is carrier-free; adding carriers negates that benefit).

Reconstitution and buffer (item-specific formulation is not provided):

  • Reconstitution buffer, concentration, and pH: Not specified for this item; refer to CoA/Spec Sheet. In general, antibodies are reconstituted or diluted in sterile PBS or TBS at neutral pH.

Stability notes (general):

  • Avoid vigorous mixing/foaming; gently invert to mix.
  • Do not store working dilutions for extended periods unless stabilized and sterile-filtered.
  • Record freeze–thaw history; discard aliquots showing precipitation or loss of activity.

For definitive guidance on concentration, excipients, and shelf life, consult the CoA/Spec Sheet.
