Lithium carbonate (Li₂CO₃) can serve as a catalyst in certain chemical reactions and is also clinically used for its antimanic effects. The bluing step after HE staining or other tissue staining is very important. Hematoxylin appears red under acidic conditions (in its red ionic state) and blue under alkaline conditions (in its blue ionic state). After differentiation with hydrochloric acid-ethanol, tissue sections appear red or pink. Therefore, immediately after differentiation, water is used to remove the acid from the tissue sections to stop the differentiation process. Then, a weakly alkaline solution is applied to make the hematoxylin-stained nuclei appear blue. This process is called bluing or blueing.
Lithium carbonate aqueous solution (1%) is mainly composed of lithium carbonate and deionized water. It is frequently used for bluing after HE staining or other tissue staining and serves as a very important auxiliary reagent.
Protocol (for reference only):
1.Follow the specific experimental requirements. Rinse thoroughly with water immediately after bluing.
2.The bluing time generally ranges from 3 to 30 seconds, depending on the specific tissue type and section thickness. Adjust the time as needed.
Precautions:
1.Keep the container tightly sealed. Use promptly after opening, as the active components are volatile.
2.For your safety and health, please wear a lab coat and disposable gloves during operation.
3.For research use only. Not for clinical use.
