The purpose of fixation is to preserve the original morphological structure of cells and tissues. Fixatives can prevent autolysis of tissues and cells by endogenous lysosomal enzymes, inhibit the growth of bacteria and fungi, and inactivate enzymes by altering the internal structure of proteins through coagulation, formation of addition compounds, etc. Fixatives are mainly divided into aldehyde fixatives, mercuric fixatives, alcohol fixatives, oxidizing agent fixatives, picrate fixatives, etc. The more commonly used ones are formalin among aldehydes and ethanol among alcohols.
Metabolic Bone Disease Fixative is similar to neutral formalin fixative and is mainly composed of formaldehyde, phosphate, and deionized water, with a pH of approximately 7.35. This fixative is especially suitable for the fixation of metabolic bone disease tissues.
This reagent is for research use only and is not intended for clinical diagnosis or other purposes.
Procedure (for reference only):
1.Operate according to the specific requirements of the experiment. Generally, the fixation time for specimens should be controlled at 1–4 h/mm; for large specimens, the fixation time should be appropriately extended.
2.After fixation, tissues can be stored long-term in neutral formalin fixative (10%) or 70% ethanol.
Precautions:
1.Metabolic Bone Disease Fixative is somewhat irritating and corrosive. Please operate carefully in a well-ventilated environment and avoid inhalation.
2.The fixation time varies with the thickness of the tissue sample. The appropriate thickness for routine biopsy tissues is 2–4 mm, generally not exceeding 6 mm; proper selection of tissue is conducive to the penetration of the fixative.
3.The volume of fixative should be sufficient. Generally, the volume ratio of fixative to tissue block should be greater than 10:1; if the volume is insufficient, the fixative can be replaced 1–3 times during fixation.
4.Temperature has a significant effect on fixation. Increasing the temperature can accelerate fixation, but the temperature should not be too high.
5.After removing fresh tissue, it should be fixed promptly. If timely fixation is not possible, it should be stored in physiological saline and sent for examination promptly.
6.For your safety and health, please wear a lab coat and disposable gloves during operation.
7.After opening the reagent, please use it as soon as possible to prevent adverse effects on subsequent experimental results.
