Panoramica

NADPH oxidase (NAO) is a typical membrane protein that catalyzes the oxidation of NADPH to NADP⁺ while transferring electrons to oxygen to generate superoxide anions. It is widely distributed in animals, plants, and fungi. Dysfunction of this enzyme can lead to chronic granulomatous disease (CGD) in humans, while in plants, it is closely associated with disease resistance and various stress responses. NADPH oxidase (NAO) converts NADPH to NADP⁺ and simultaneously generates superoxide anions (O₂⁻), which subsequently react with a chromogenic agent to form a water-soluble yellow product. Background values are excluded by adding the specific inhibitor diphenyleneiodonium (DPI) as a control. The activity of NAO is calculated by measuring the absorbance of the resulting colored product at 450 nm.
