Panoramica
Phalloidin is a bicyclic heptapeptide toxin that binds specifically to the interface between Factin subunits, locking adjacent subunits together. Compared with actin monomers, phalloidin binds much more strongly to actin filaments. It stabilizes actin filaments by decreasing the rate constant for dissociation of actin subunits from filament ends, thereby effectively inhibiting filament depolymerization. In addition, studies have shown that phalloidin can also inhibit the ATP hydrolytic activity of Factin. The toxin stabilizes the Factin structure by trapping actin monomers in a conformation distinct from that of Gactin and greatly reducing the monomer dissociation rate constant associated with ADP trapping.
The cellular effects of phalloidin are concentrationdependent. At low concentrations, it recruits weakly polymerized actin and filamin in the cytoplasm to form stable actin polymer “islands” without disturbing the structure of stress fibers (i.e., thick bundles of microfilaments). This property makes it an important tool for studying the distribution of Factin: fluorescently labeled phalloidin derivatives can stain actin filaments for optical microscopy. Fluorescent phalloidin derivatives enable precise localization of actin filaments in both live and fixed cells, and can also be used for the visualization of single actin filaments in vitro, serving as a key tool for highresolution studies of actin networks.
Product Advantages
High binding specificity: Selectively binds polymeric Factin (fibrous actin) with little to no binding to monomeric Gactin, resulting in a high signaltonoise ratio.
